A spokesman from the Ambulance Service of NSW said it was believed the vehicle rolled over on the Hume Highway at Tarcutta.
Two passengers died at the scene, two have been airlifted to hospitals in Melbourne and Canberra and the other three patients, suffering lacerations, have been taken by road to Wagga Base Hospital.
The fatalities are the first on Border roads this holiday period.
Southbound motorists are being diverted around the closure using the Sturt Highway via Wagga Wagga, then the Olympic Highway to Henty and Culcairn continuing back to the Hume Highway north of Albury.
Northbound lanes of the Hume Highway remain open and unaffected.
Southbound lanes are expected to remain closed for several hours to allow NSW Police to investigate the cause into the fatal incident.
Motorists travelling southbound via this location are urged to following diversions, exercise caution and allow additional travel time.
