Albury police Inspector David Cottee said the car was travelling north when it flipped and rolled near the Borella Road overpass some time between 2.30am and 2.45am today.
Three people have been taken to hospital, including a female passenger who is believed to be pregnant.
The male driver is being treated for cuts to his head while another male passenger is being treated for severe cuts to his leg and face.
Inspector Cottee said speed and alcohol were both believed to be factors in the crash.
Police have closed the northbound on-ramp to the Hume Highway at Borella Road and traffic has been reduced to one lane beneath the overpass.
Police are asking motorists exercise care in the area as they set up a forensic investigation.
