More than 200 classic trucks will make the journey up the Old Hume Highway, starting at the Ford factory in Melbourne and cruising north.
The trucks will stop at Winton for a lunch break from 11am to 1pm before progressing to Glenrowan, Wangaratta, Barnawartha and finally Albury.
Brands of trucks taking part in the cruise include International, Kenworth, Mack, Commer, Dodge and other well-recognised names from trucking history.
Spectators will be admitted to Winton for a gold coin donation, with proceeds to be donated to Trans-help, a volunteer organisation supporting families and people in the transport industry.
