The Nathalia man, aged in his 40s, was driving on Pinnucks Road near the Goulburn Valley Highway when his vehicle left the road and struck a tree about 12.20pm.
Emergency services attended the scene but the man could not be revived.
Police are investigating what led to the crash.
Meanwhile, two men died in a three-car smash near Melbourne’s Tullamarine airport.
The crash occurred just after 5pm yesterday on Sunbury Road when the driver of a blue Holden lost control of his vehicle.
The Holden collided with an oncoming car and another car following it.
“Two male occupants in a second vehicle were killed,” a Victoria Police statement said.
“The driver of the Holden was taken to The Royal Melbourne Hospital with serious injuries.”
The male driver of the third vehicle was not injured.
The deaths took Victoria’s road toll to 142.
