The 15-year-old Richmond boy died on Saturday night when the vehicle crashed into a tree off the Murray Valley Highway.
He was rushed to Wangaratta Hospital but died shortly after arriving.
His 17-year-old brother was getting driving experience on his learner permit and was being supervised by their father.
An alcohol breath test after the crash returned a negative result.
Investigators from the Major Collision Investigation Unit are working to determine a cause but speed does not appear to have been involved.
The Melbourne-based officers will work with local police to determine why the Ford sedan left the road.
The vehicle, which had been travelling west before the crash, was towed from the scene.
The incident occurred about 9.15pm.
The two passengers received minor injuries.
