The ambulance helicopter is on the scene of the Rutherglen-Springhurst Road to crash to fly two of the victims to the Royal Melbourne Hospital.
Paramedics were called to the collision just after 6.30am.
A 23-year-old woman has serious head injuries and is in a critical condition.
She was trapped for just over an hour.
A 19-year-old man has facial and leg injuries and is in a stable condition.
He was trapped for more than 90 minutes.
The man and woman were treated by paramedics throughout the rescue operation.
A woman from the third car has been assessed by paramedics and didn't require any treatment.
Police from the Major Collision Investigation Unit are on their way from Melbourne.
UPDATE 8.50am: Air ambulance has arrived on scene.
North East drivers should avoid using Rutherglen-Springhurst Road this morning as emergency services respond to a multi-vehicle collision.
An accident occurred around 6.30am this morning near McPherson’s road north of Springhurst and south of Rutherglen.
VicRoads advises McPhersons Road is closed and for motorists to detour via Chiltern-Rutherglen Road.
