It is believed a car was travelling northbound on the Hume Freeway about 2am when it collided with a cow on the road.
After the four occupants had stepped out of their car, a second car travelling in the same direction collided with the cow and struck two people.
A man aged in his 50s, who was a passenger in the first car, died at the scene.
A man aged in his 40s, who was also a passenger in the first car, was transported to hospital with non life threatening injuries.
No one else was injured in the incident.
Diversions are currently in place near the Murray Valley Highway and police are asking people to avoid the area if possible.
