The 58-year-old woman, who was driving alone, was travelling from Beechworth to Wodonga wen she came off the road near Magpie Lane at Wooragee.
Beechworth Leading Senior Constable Andrew Deegan said police were called to the scene about 6.15pm on Wednesday.
“She drove off the road towards the left, became airborne and the car landed on its roof,” she said.
“She had to be extracted by CFA and SES, it took about two hours.
“She was transported to Albury Base Hospital with serious but non life-threatening injuries.”
Leading Senior Constable Deegan said the accident was not the result of road conditions and said it was cause to remind people to respect that stretch of road.
“The cause is not likely to be road, environmental or fault of the car … there was no reason to have an accident,” he said.
$9.6m was committed earlier this month to improve safety on the road, including introducing an 80km/h speed limit through Wooragee.
