Mackinlay started his run by having Jamie Cooper caught behind before bowling Steve Brand, Vince Chaffey and Layne Heinjus.
He finished with career best figures of 6-23 off eight overs and is only the fifth HDCA player to claim a double hat-trick.
“I got a bit lucky,” Mackinlay, 19, said.
“For some reason I swung the ball a bit which doesn’t usually happen.”
Mackinlay’s previous best figures were 5-25 against Culcairn last season.
But his afternoon didn’t finish there.
With Culcairn making 7-137 thanks to good knocks from Rory Jenkins (47) and Eddie Turner (41), Mackinlay made 67 not out in the chase with Holbrook winning with just over two overs to spare.
Hayden Wishart provided good support with 48.
Osborne comfortably defeated TRYC.
