Constable Dimity Hinch, of Wodonga police, said the truck, which was carrying household goods, drifted onto the median strip before rolling about 2.30am.
The crash happened about 10 kilometres south of Wodonga in the north-bound lanes.
Police are investigating the cause of the crash but say speed was not a factor.
Both north-bound lanes are expected to be closed until later this morning. Diversions are in place.
Northbound traffic is being diverted onto the Murray Valley Hwy along the Old Barnawatha Road and back onto the Hume Fwy.
Emergency services are on site directing traffic. Motorists are advised to allow extra time for their journey.
