Police said the man was driving a Holden sedan on the Wangaratta-Kilfeera Road when he lost control and crashed into a tree at 3.20pm.
He died at the scene.
There was no one else in the car at the time.
Early investigations indicated distraction, but not speed, may have been a factor in the crash, police said.
A passer-by is believed to have called Triple 0.
Police urged anyone that witnessed the crash to phone Wangaratta police on (03) 57230 888.
The death has come on the same day Victoria Police assistant commissioner Robert Hill launched the "Road to Zero", a new campaign to encourage the community to help reduce road trauma.
Sixty-eight people have died on Victorian roads this year; seven more people than last year.
