Firefighters were called to the Greta Road house at Wangaratta about 11.20pm on Monday.
A fault in a chimney flue appeared to have caused smoke to spread into the roof cavity.
Leading firefighter Murray Walker said the occupants were freeing children from the house when fire crews arrived.
They removed a section of the roof to bring the incident under control.
