Witnesses told police the Winnebago left the road gradually before colliding with a tree.
The driver was trapped for a time, with SES and CFA officers working to free her, but she died at the scene.
The speed limit along that particular stretch of road is 100 km/h.
Sergeant Michael Connors from the Wangaratta Highway Patrol said the woman was a local, travelling from Bright to Myrtleford.
He ruled out excessive speed as the cause of the crash.
“For some unexplained reason, the vehicle has left the road gradually, and not at a high speed, and collided with a tree,” he said.
“There were no other vehicles involved.
“A 60-year-old female from the area has died at the scene.”
The investigation into the crash is ongoing, Sergeant Connors said.
“We’re looking at a number of avenues and have a number of witnesses to the collision,” he said.
“We can definitely rule out speed, but we are looking at other fatigue issues.
“We’ll also look at the vehicle to see if there are any faults there.
“We’ll look at the driver’s medical history as well, we’ll take into account witness statements and collate it all for the coroner.”
Sergeant Connors said the road fatality was a tragedy for the woman’s family and community.
“We’re talking about locals dying on local roads, this is a prime example of that,” he said.
“Any fatality is devastating, for this family, they’ve lost a loved one in circumstances that probably could have been avoided.
“We had a lot of SES, CFA and ambulance officers at the scene, the driver was trapped in the vehicle for some time – it was a fairly lengthy process to remove her.”
The fatal crash brought the Victorian road toll to 110, 23 fewer than at this time in 2017.
