McDonald, 40, took out the three-day 10km swim and 150km bike ride (day one), 275km bike ride (day two) and 84km run (day three) in a record time.
Forty-eight competitors took part in the invitational event.
McDonald has been based in the US for about six years and is a regular winner on the ironman circuit.
“I was attracted to Ultraman Florida after my teammate Jamie (Harris) won the event last year and talked very highly of it,” McDonald said.
“I have also enjoyed visiting Florida a lot and it's a perfect chance to see 320-plus miles a little closer."
McDonald has previously won back-to-back Louisville ultra-marathons.
His father, Neil, and brothers, David and Daniel, have also been heavily involved in the sport.
