Emergency services were called to the freeway between Winton and Glenrowan about 11.15pm on Wednesday.
A Drysdale man, 54, was travelling north in a 1964 ex-army amphibious vehicle at about 80km/h when he was hit from behind.
The impact caused the vehicle to roll down an embankment and eject him through a hatch.
Police struggled to find the vehicle due to darkness.

They eventually found the man about 10 metres from the vehicle after he shouted out for help.
He was airlifted to The Alfred Hospital in a critical condition.
The truck driver has been spoken to by police and released pending further enquiries.
