Spencer, who has played 26 senior matches for the Roos, is one of the youngest players in the 43-member squad at 19.
"It's well deserved," Roo coach Marc Almond said.
"He's earned it.
"Tyler had a good pre-season and is wanting to improve his game each time he plays.
"He's a great kid, very coachable."
Ladder leader Wangaratta dominate the training squad with nine players - Jamie Anderson, Michael Bordignon, Ben Douthie, Matthew Grossman, Matt Kelly, Brad Melville, Luke Morgan, Michael Newton, Josh Porter, Joe Richards and Ben Speight - in the mix.
Co-coach Brad Murray said he was pleased with the squad, with top-liners including Panther Shaun Mannagh keen to play if free of VFL commitments.
"It's a pretty good even mix of key talls, inside mids and outside mids," Murray said.
"Playing in Melbourne you would think the grounds will be slightly smaller so we will try and make sure we have good grunt in there.
"Hopefully we get through the next few weeks and put out a good side."
Morris medallist Brodie Filo is the most notable player missing after he spent the off-season playing in NT.
The match will be played on May 18.
