About 60 people were evacuated from the Smollett Street property on Tuesday morning after a letter containing the green powder was opened about 8am.
It had been in an envelope in the mail room, and got stuck in a sorting machine.
It was released when the staff tried to dislodge it from the machine and the three female staff were decontaminated.
The mail room was secured and the level evacuated, and the powder recovered by police.
"I can report there were no injuries, the three employees that were in the mail sorting room at the time did not appear to be affected by the substance and were seen at the scene by the NSW Ambulance," Inspector Scott Russell said.
"That substance is in police possession and it will go to our laboratory for further analysis to determine what substance that is."
He said there hadn't been any threats made, and staff handled the situation well.
IN OTHER NEWS:
Police could know what the substance is by Wednesday afternoon.
Fire and Rescue NSW Superintendent Stewart Alexander said "we don't take any chances with these types of incidents".
"We took every precaution to make sure people in the vicinity of the mail room were well looked after," he said.
Smollett Street was blocked to traffic for about two hours.
