Emergency services were called to the incident on the Great Alpine Road about 5.20pm on Thursday.
The Myrtleford woman's car hit at tree about 70km/h near the town and she was flown to hospital.
Acting Sergeant Patrick Murray said the vehicle, which had been travelling east, was extensively damaged with all airbags deployed.
"Due to the safety design of the car, she's still alive," he said.
"That's why the ANCAP safety rating is important when buying a car.
"At this stage, it's unknown why she left the road.
"We're looking at all avenues of enquiry as to why this has occurred."
