The St Pats captain, 19, finished with a mammoth total of 39 votes, eclipsing the 35 she polled when breaking through for the association's top honour in 2016.
Mahoney was 16 clear of runner-up Bridget McDiarmid, the winner of the past two Star Player awards for Albury City, with Melrose's Maya Davis third with 17 votes.
Wangaratta duo Bianca Mulqueen (16 votes) and Leanne Branson (15 votes) rounded out the top five.
Mahoney was thrilled to be crowned the league's best and fairest player once more.
"Especially with so many incredible players in the league, I feel very grateful to get the votes off the referees. It means a lot," she said.
"I'm very happy here and I love the club, that's why I've never gone (to play at a higher level) previously."

She also finished as the league's leading goal-scorer with 21, edging out Albury United's Alicia Torcaso and Mulqueen, who booted 19 apiece.
Mahoney will look to cap off the perfect season when she leads her side into Sunday's cup final against Wangaratta at Jelbart Park.
St Pats are going for the AWFA treble after winning the league title and the Iannotta Cup pre-season competition already in 2019.
"We've trained really hard this week and I think it's the most important game of the year, so it would be great to get the result," Mahoney said.
"We just want to really enjoy the game and put all our effort from the year and the pre-season into it."
The Star Goalkeeper award also went to St Pats with Natasha Anderson picking up 43 votes to beat last year's winner, Melrose's Rachael Reitano, by one.
"I got pipped at the post by a couple of points, but I came back this year and was determined to win it," she said.
"It was unfortunate that Rachael was unable to play the last few games because of her broken finger, but kudos to her, she's a really great goalkeeper and I'm glad to be up against someone of her quality."
Twin City's Chantal Dunphy and Myrtleford's Kayla Kneebone tied for the reserve women's Star Player on 27 votes.
Kneebone also finished as the leading goal-scorer with 26, while Nikita Tabuteau-Rosas continued a big night for St Pats by claiming the reserve Star Goalkeeper award with 36 votes.
St Pats' Mike Fruend and Albury United's Michelle McGrath were named Volunteer of the Year winners, United's Riley Semmens and Hotspurs' Lily Godfrey shared the Junior Club Person of the Year, while Hotspurs' mentor Fiona Smith was Coach of the Year.
