Police said the driver of the boat, a 35-year-old Wodonga man, had been interviewed and released pending further inquiries.
"It is believed a ski boat was pulling a small tube carrying two people when they were thrown off and hit a tree about 4.20pm," a police spokeswoman said.
"A 21-year-old Wodonga woman was airlifted to hospital with life-threatening injuries and an 18-year-old Lavington man was hospitalised with serious injuries."

The Alfred Hospital said on Monday afternoon the woman's condition was critical.
EARLIER: A woman was critically injured in a boating accident in Ebden on Sunday afternoon.
Ambulance Victoria said paramedics were called just before 4.30pm.
"A woman in her 20s suffered a head injury and was airlifted to the Alfred Hospital in a critical condition," Ambulance Victoria said.
"A male in his late teens has suffered leg and hip injuries and was transported by road to Albury Base Hospital in a stable condition."
Victoria Police said the incident occurred at Ludlows Reserve, Murray Valley Highway, at 4.25pm but could not release any further details at this stage.
