Wangaratta Highway Patrol officers intercepted a Kia station wagon at Glenrowan while conducting speed checks on Monday morning.
The car was detected at 137km/h in a 110km/h zone and the female driver, 39, returned a positive result to a drug test.
"Police inspected the vehicle and located an unrestrained four-year-old child lying on the back seat and a 10-year-old child lying under a doona in the rear compartment of the car," a police spokeswoman said.
"The Thomastown woman will be charged by summons with excessive speed, impaired driving, drive whilst suspended and unrestrained children."

The incident occurred near the end of Operation Regal for the long weekend.
Wangaratta Senior Sergeant Darren Wittingslow said 21 drink and drug drivers were caught with 341 offences detected.
Three dirt bike riders crashed and police attended a separate smash in Yackandandah, but nobody was seriously hurt.
"Overall driver behaviour was good, with large volumes of traffic over the weekend," he said.
Albury officers also intercepted an unlicensed driver on Mate Street on Monday night.
The 22-year-old was intercepted about 10.15pm but was again stopped a short time later while driving and returned a blood alcohol reading of 0.108.
He was charged with unlicensed and mid-range driving and will front Albury court on August 3.
Detective Acting Inspector Chris Wallace said police had issued 141 infringement notices in the Albury region during the operation.
