McCullum was approved to train by Racing NSW earlier this month.
The 29-year-old is in the process of purchasing the stables owned by Brett Cavanough and most recently used by Mitch Beer.
McCullum said she had been passionate about racing since she was a teenager and was proud to be joining the Border training ranks.
"It was something I initially started five years ago but put on hold because the timing wasn't right," McCullum said.
"But I have now been able to achieve my goal and have been given a golden opportunity with the support of some owners."
McCullum recently married jockey Scott McCullum and was previously an amateur jockey under her maiden name Laura Collins.
The pair have two children.
McCullum also worked under Brian Cox as a teenager for more than three years.
"Brian taught me a lot while I was working for him," she said.
"I've also had stints with other trainers in Victoria, NSW and Queensland.
"I was only an amateur jockey for two years because of injury as well as taking time off to have the kids."

McCullum will initially train a dozen horses.
"We have signed all the contracts and will move into Brett Cavanough's former stables," she said.
"To start off with Racing NSW has restricted me to training 12 horses.
"That will be reviewed after three months and depending how the team has performed I intend to expand on that."
McCullum is hopeful of having her first runner in two weeks.
"I won't be able to have a runner before that because I have to do two weeks isolation because I have been working in Wodonga," she said.
"I intended to have a runner at Albury's meeting on Monday but I will have to wait until July now."
McCullum said it wouldn't have been possible to gain her trainers licence without the support of Cloverlea Farm.
"Cloverlea Farm bred all the horses that I will be training," she said.
"Their support has been amazing and I wouldn't have been able to get this far without it.
"My partner Scott has also supported me along the way and helped make this all possible."
Meanwhile Albury Racing Club will host it next meeting on Monday.
The club has received 133 nominations for the seven race card.
Mitch Beer looks set to heavily target the meeting with seven nominations.
Wangaratta Turf Club will also race on Monday.
The club received a whopping 600 nominations for its eight race card.
