Officers have been told a 26-year-old woman attended a party at a house in Yarraville, near Footscray, in September 2016.
About 15 people were at the gathering, celebrating the senior club's premiership.
The event was not sanctioned by the club, which police have chosen not to name for privacy reasons.
The woman complained of feeling unwell after drinking a glass of orange juice and was taken to a bedroom.
She woke the next day with injuries consistent with being sexually assaulted and immediately left.
The assault was reported to police in December 2018 with the matter still under investigation.
Detective Senior Constable Adam Vaughan on Thursday told The Border Mail there would be people with information who could assist with the investigation, and urged them to come forward.
"Any information is relevant," he said.
"I'd encourage people who have information, whether they were there or heard it from other people who may have been there, to contact police or Crime Stoppers.
IN OTHER NEWS:
"If you were at a residence in Yarraville and hosting a party celebrating a grand final win in North East Victoria, people would know the incident that we're talking about."
Detective Constable Vaughan said the woman had been shaken by the ordeal.
"The victim was quite traumatised at the time," he said.
"It's obviously had a huge impact on her life and she's in a space now where she was comfortable to disclose to police what occurred.
"We're trying to get as much information as possible so we can get a clearer picture of what happened that night."
No-one has been questioned or charged over the incident.
Detective Senior Constable Vaughan said people could attend their local police station or anonymously call 1800 333 000.
