Victoria Police said the incident occurred after the men were picked up at a property in Victoria Parade, East Melbourne about 12.30am.
"During the journey it's alleged one of the men asked the driver to head to another address," a police spokesman said.
"The driver declined and the man assaulted the driver."
The victim stopped his car and got out to call police and the offender moved into the front seat, attempting to drive off.
Unsuccessful, he and his friend ran from the car.
Police arrived and arrested a 29-year-old man from South Melbourne and a 28-year-old man from Yarrawonga, who are both assisting police with their inquires.
