Sir Angus Houston, who was appointed last month, told this week's Murray Darling Association national conference he would be open and transparent in the role.
"I will be collaborative and inclusive," he said.
"I will be impartial and show respect."
Sir Angus said he accepted the "tough gig" because "quite simply, having the Basin on track is critical for our nation".
"The Murray-Darling Basin is an ecological and economic powerhouse," he said.
"It is about the nation's economy, it's about the wellbeing and the future of First Nations people, and the health of farming communities that clothe us, feed us, and care for the country.
"I will do my best to be part of the solution to water policy across communities and governments.
"To give the rivers what they need and strong communities what they need."
