The "inconclusive" detection was on January 11, but DHHS confirmed yesterday it was no cause for concern.
"Wastewater monitoring is continuing in more than 60 locations around the state," a spokesperson said.
"An inconclusive wastewater result is a result which is not able to be independently confirmed.
"After an inconclusive result, as well as any positive detection, the frequency of wastewater monitoring is increased.
"Further samples have been taken since 14 January that did not detect viral fragments and additional sampling is ongoing."
It was the 13th consecutive day of no community transmission in Victoria, with four new cases overnight, all in hotel quarantine.
One of those cases is a returned traveller but the other three are related to the Australian Open.
Premier Daniel Andrews responded to concerns from mayors of Sydney local government areas whose localities remain in "the red" despite no COVID-19 cases.
"I'm not particularly interested in providing messages to mayors in other states - they've got a job to do, so do I, and that's following the public health advice," he said.
"As soon as we can release those local government areas ... we absolutely will."

Mr Andrews said Brisbane turning 'green' soon, allowing Queenslanders free entry to his state with a permit, was "a real chance".
"Hopefully we can have NSW green very soon as well," he said.
Mr Andrews said he was "more than happy" to look at a permit for tourism purposes in areas of low risk.
"The declaration of green last night ... should deal with that," he said.
"We're always looking at sectors that have been hit very hard, and we would never rule out doing more.
"I'm not announcing a package today, but we'll look at [it] as each month rolls on."
