The Saints headed into the clash at McNamara Reserve without goal shooter Amie Antonello, with Rebecca Piazza stepping in to fill her spot.
Piazza shot 26 goals in the 48-32 win, while goal attack Grace McKereal was a standout for the Saints.
Defender Rhiannon Mcilroy made the step up from B-grade to aid the side.
"They adjusted to the changes really well," Myrtleford coach Tina Way said.
After consecutive losses to top five sides Yarrawonga, Wangaratta and Lavington, Way admitted the Saints were hungry to add to their first round win against North Albury.
"We had three losses in a row and we really wanted to get a win on the board," she said.
"We've had a few poor starts against the top sides, so we focused during the week on starting our game well."
Way also had a strong game in defence and is finding the balance between being a player and coach.
"Each week I'm getting to know the girls better on court and that makes being a playing coach easier," she said.
Brooke Pryse and Mackensey House both had solid games for the Raiders, while new goal-shooter Taylor Donelan landed 19 goals.
In other matches, North Albury got over the line in a thriller against Wodonga at Martin Park.
The Hoppers were trailing at every break before a 10 goal final quarter saw them ahead by one.
Natalie Heagney shot 26 goals, while Emily Browne was a standout for the visitors.
Maddie McFarlane and Sarah Cardwell were the best for the Bulldogs.

Wangaratta bounced back with a 19-goal win against Albury.
Amanda Umanski finished the game with 38 goals, while Issy Byrne worked tirelessly in centre.
Heidi Fisher and Gabrielle Mcrae were best on court for the Tigers.
Yarrawonga was too good for Wangaratta Rovers at W.J Findlay Oval, with the Pigeons securing the win 50-36.
Laura Davis and Bridget Cassar were the standouts for the victors, while Katey Pryor and Amelia Mathewson were the best for the Rovers.
Corowa-Rutherglen joins Yarrawonga in being undefeated this season after a 25-goal win against Lavington.
Jennifer Barratt and Sophie Hanrahan were strong in defence, as was Sarah Meredith for the Panthers.
Yarrawonga, Corowa-Rutherglen, Wangaratta, Lavington and Myrtleford make up the top five.
