Officers will start a four day blitz on Friday, with automatic number plate recognition technology to alert members to Melbourne region number plates.
City residents can only travel 25 kilometres from their homes.
Senior Sergeant Darren Wittingslow said while there would be less vehicles on the road due to the city restrictions, the Queens Birthday weekend was a dangerous time on the road.
"It's a risky weekend with extra road users," he said.
"We want people to drive to the conditions and ensure they don't speed, don't get distracted and wear seatbelts.

"We'll be doing preliminary breath tests and preliminary drug testing for drivers across the weekend."
In the five year period to 2019, there were two deaths, seven people seriously injured and 12 people who sustained minor injuries in the Wangaratta and Alpine regions during the long weekend.
Senior Sergeant Wittingslow said highway patrol and general duties police would be enforcing the rules.
He said police cars with number plate recognition systems received an alert when Melbourne vehicles were detected.
There have been few Melbourne region residents detected in the North East since the restrictions came into force.
"It will be a bit different this year with only regional people traveling on our roads, but there is still that heightened risk of road trauma," Senior Sergeant Wittingslow said.
