Fifty people are already booked in to be vaccinated at Beechworth Pharmacy, owned by Ruth Parker.
With no GPs in the area opting to participate in the vaccine roll-out, Ms Parker was keen to get the pharmacy involved.
"For me, submitting the expression of interest to the government was really important because we were getting a sense that there was a bit of a gap in the community," she said.
"We just saw that the doctors are so overloaded with all the chronic illnesses that have sort of been left behind in COVID.
"We have a lot of elderly customers and they were asking us about access - I think they were getting a bit scared that they wouldn't be able to access it in a timely manner.
"Some of them don't leave the community at all, they rely on the services available here.
"We did some renovation work at our own pharmacy to hopefully be successful in that tender and we were notified two weeks ago that we were successful, which is really great.
"We're excited to get started on Monday."
Ms Parker said the pharmacy was limited to vaccinating only people over the age of 60 with AstraZeneca and hoped they could do Pfizer down the track.

"I'm hopeful we can do it as well - it would be possible for us to do it, if we had the right storage given to us," she said.
"People are definitely asking about the Pfizer."
Yackandandah is also vaccinating with AstraZeneca.
