A Balnarring man, 28, and a Frankston man, 36, were charged with trafficking a commercial quantity of a drug of dependence.
The pair were also issued with fines for breaching the Chief Health Officer's directions.
Both men have been remanded and will appear in the Wodonga Magistrates Court on October 14.
The vehicle, a tow truck with Victorian registration plates, was intercepted by officers performing roving patrols on the Hume Highway near the border as part of Operation Sentinel just after midnight.
Detectives from the Wodonga Criminal Investigation Unit seized the tow truck and another vehicle on the back of the truck.
MORE NEWS:
A subsequent forensic examination confirmed the substance concealed within the vehicle was more than 15kg of methylamphetamine, estimated to be worth up to $10 million street value.
Wodonga Criminal Investigation Unit, Senior Sergeant Garry Barton said this was "a terrific piece of policing by our local members".
"While the border patrols taking place as part of Operation Sentinel are playing a crucial role in keeping those from New South Wales without valid permits out of Victoria, they also allow our members to more easily identify and stop serious criminal behaviour such as this," he said.
"This is a significant drug bust, as we know the harm substances such as methylamphetamine cause to our communities.
"We will continue to ruthlessly target those looking to profit from drug transactions, which have severe consequences from a crime and public health perspective."
EARLIER: Police discovered about 15 kilograms of suspected methamphetamine during a routine border traffic check at Wodonga on Tuesday.
Detective Senior Constable Brianna Gibson, of Wodonga Crime Investigation Unit, said police intercepted a vehicle on Hume Freeway near the Melrose Drive off ramp in the early hours.
A 2011 blue Ford DC ute was being transported on a tow truck.
"It was identified that the vehicle and the occupants were returning from Sydney, entering Victoria and breaching COVID restrictions," Detective Senior Constable Gibson said.
A search located the drugs. Their value is still being established "but I can assure you it would probably be within the millions".
IN OTHER NEWS:
The detective said it was too early in the investigation to release further details about the driver and any passengers.
Inquiries continued into the source of the drugs.
"It is concerning to have a large amount of drugs on the street, but we're more than happy with this result to get this amount off the street and protect the community," she said.
