He returns home after a three-year stint at Lavington.
Mackinlay enjoyed a breakout season in 2019 when part of the Panthers drought breaking flag against Wangaratta.
The dangerous forward played the maximum possible of 35 matches with the Panthers and booted 41 goals.
Mackinlay said he was looking forward to the challenge and extra responsibility of being assistant coach under Matt Sharp.

"My priorities in regards to what I want out of my football career have changed a bit," Mackinlay said.
"When I initially joined Lavington I wanted to test myself and play at the highest level I could.
"But now I want to have success with my home club which is the major reason behind my decision.
"I feel having three seasons at Lavington has been good for my development and playing under a coach the calibre of Simon Curtis.
"Graham Hart has also been good from my development and playing alongside guys like Tom Hargreave, Marty Brennan and Luke Garland who lead by example."
Mackinlay's father, Robbie, is the all-time games record holder at the club after 369 matches with the Brookers.
He was also a dual premiership player and Barton medallist in 1994.
"I think I previously played 115 matches with Holbrook before I joined Lavington," he said.
"I joke with dad that I've only got about a dozen more seasons of injury free football to go to give his record a shake."
Mackinlay's arrival will help narrow the gap between the Brookers and Osborne with the minor premiers undefeated from 16-matches.
The Brookers lost both encounters to the Tigers this season by 51 and 66 points.
"Osborne is clearly a class above every side in the competition at the minute," he said.
"It's going to be a steep hill to climb to bridge the gap but we'll see what we can do.
"The exciting part is the upside with the kids on our list like Corey McCarthy, the two Holmes' boys (Lachie and Will) and Ewan Mackinlay.
"I think that's where we will get the most improvement on our list.
"Whether or not we add a few more recruits remains to be seen and would obviously help the cause.
"But Sharpie has pinned a lot of faith in the kids since arriving as coach and is starting to reap the dividends now."
