COVID Commander Jeroen Weimar said today was a significant set back.
He said if the trend continues it would have grave implications for the health system.
"This outbreak still remains within our hand, we still have lots left to play for," he said.
Mr Weimar said the jump was a direct consequence of hundreds of decisions made over the long weekend.
He said there were 46 cases in regional Victoria including five new cases in Shepparton.

Victorian Premier Daniel Andrews said today's huge spike in case numbers marked a 50 per cent rise on yesterday's case numbers.
He said it would be a 'great concern' to Victorians.
Mr Andrews said the state knew case numbers would increase but they increase even more when people don't do the right thing.
He said coming off a long weekend, many of the cases were completely avoidable.
The Pfizer interval will be halved in state hubs, from six weeks to three weeks.
The new cases brings the total for Wodonga to six.

Victoria's daily case numbers have spiked drastically with 1438 cases and five deaths recorded in the 24 hours to midnight.
The new record eclipses the previous record highest daily cases set yesterday.
In the 24 hours to midnight 34,323 vaccines were administered and 65,497 tests were received.

It comes as the Victorian Health Department faces more than $95 million in fines after it was charged over hotel quarantine failures.
A filing hearing in Melbourne Magistrates Court is set for October 22.
Seventeen of the charges relate to failing to provide and maintain, as far as reasonably practicable, a working environment that was safe and without risks to health for its employees.
