Mr Quilty said he has received the COVID vaccine but will not comply with any government requirement to provide proof.
The state government intends to introduce legislation to mandate MPs have their first jab by October 22. The motion would block MPs who do not provide proof of vaccination from attending parliament until the second day of the 2022 parliamentary sitting.
Liberal leader Matthew Guy told The Age the Coalition would support the motion.
Mr Quilty said his decision was about standing with Victorians who 'do not wish to be forced into vaccination'.
"I will not play their game," he said.
"I am not an anti-vaxer. I got my vaccine as soon as I was eligible.
"I encourage everyone to get vaccinated. I think the virus is more dangerous than the vaccine for most people.
"But I refuse to participate in the immoral coercion of mandated vaccines. I will not comply."

Treasurer Tim Pallas said MPs have a moral imperative to provide their workplace with proof of vaccination, as many Victorian are required to do.
He said not complying might be an expression of the Liberal Democrat MPs' liberty but was also an 'existential threat' to those they interact with.
"People have to be sensible, this is not an ideological debate and a mask is not a political statement it is a public health imperative," he said.
"I respect the Liberal Democrats have different view and different priorities but the virus doesn't differential between their concerns. They need to get vaccinated and need to demonstrate [it]."
