With compatriots Jason Day and Matt Jones withdrawing from the race over the past 24 hours due to Zika virus fears, Scott Hend and Fraser are the highest-ranked Australians ahead of the July 11 selection cut-off.
Hend and Fraser are in 74th and 80th positions with John Senden next in 104th spot.
Countries are allowed two representatives in Brazil.
Fraser will play in the Bridgestone Invitational in the US this week while Senden has entered the Barracuda Championship.
Senden would need to win the second-tier event on the PGA Tour and hope Fraser falters to reel in the popular Border golfer.
His only other chance of catching Fraser – The Greenbrier Classic – has been abandoned due to floods in West Virginia.
Sixty players will battle it out in Rio de Janeiro from August 11 to 14.
Fraser leads the Asian Tour Order of Merit on the back of his thrilling victory in Malaysia in February.
He made ground on Hend earlier this week after being the top-ranked Australian in the European Tour's BMW International Open.
The right-hander finished equal 21st after rounds of 69-72-70-72.
Australians Adam Scott and Mark Leishman, who are ranked eight and 39 respectively, have ruled themselves out the Olympics over the past two months.
Mulwala shooter James Willett and Wangaratta swimmer Belinda Hocking have already made the Australian Olympic team.
Willett will be Australia’s sole representative in the double trap discipline while Hocking is chasing a gold medal in the 200-metre backstroke.
She won gold in the event at the Glasgow 2014 Commonwealth Games.
