Paramedics were called to Federation Way, north of Wangaratta, about 2.45am after a car ran off the road and rolled onto its roof.
The Holden Commodore was travelling south towards Wangaratta when the driver lost control around a left-hand bend.
Federation Way is lined with wire safety barriers, but Senior Constable Colin Boyle said the car drifted off the road through a gap at the Back Estcourt Road turnoff.
“He’s threaded the eye of a needle really and has rolled numerous times,” he said.
The unidentified deceased man was ejected from the car during the incident and located about 80 metres away from the damaged vehicle.
A man in his 20s was trapped for about half an hour.
The man was treated by paramedics for head and internal injuries during the rescue operation, and was taken to Wangaratta hospital in a serious condition.
The exact cause of the crash has not been determined and the Major Collision Investigation Unit was called in to investigate on Thursday morning.
“It’s a sad start to Easter weekend – two families are not having the Easter we want to enjoy,” Senior Constable Boyle said.
“It was a timely reminder of the dangers of 100km/h zones and police are urging people to be conscious of your speed, your fatigue and the road conditions.
“This crash occurred at 2.30am in the morning, it’s at night – make sure your driving is appropriate to the conditions.
Three people were also injured in a two-car crash on Cobram South Road about 1.20am.
Police arrived to find two cars had collided, with one person trapped in each vehicle.
A woman in her 20s with leg injuries was trapped for about one and a half hours and was treated by paramedics during the rescue.
She was flown by air ambulance helicopter to Royal Melbourne Hospital in a critical condition.
Another woman in her teens from the same car had back and spinal injuries and was treated by paramedics before being taken to Goulburn Valley Hospital in a serious condition.
A man aged in his 20s from the other car had leg injuries and was taken to Goulburn Valley Hospital in a stable condition.
Emergency crews from CFA, SES, police worked together on Wednesday evening to free the driver of another vehicle, following a rollover on the Old Hume Highway between Wangaratta and Glenrowan.
