Gabrielle Maina, who was formerly from Armidale in northern NSW, died from a single bullet wound to the neck last Thursday as she walked along a road near her home in Nairobi.
Her husband Cyrus Bernard Maina Njuguna and another man John Njuguna Waithira were arrested by police about 6pm AEST.
Police reportedly seized Mr Maina's mobile phone.
Her lawyer George Kingori told ABC earlier this week there were signs the teacher feared for her life and was forced to kneel on the road before she was shot.
It has also been revealed that Ms Maina was granted a restraining order against her former husband before her death.
The couple married in 2001 and has two sons, who are believed to be Australian citizens.
Ms Maina was a former Armidale High School and University of New England student.
Close friend and UNE lecturer Anna Cavallaro told Fairfax Media last week her death was a "tragic loss to humanity".
"I'm not exaggerating because she was one of those people that you can't forget," she said.
Ms Maina was living at Wright College in Armidale and met her husband, who is from Kenya, at university.
In 2015 she moved to Kenya to work as a teacher.
"She always wanted to help students in disadvantaged schools," Ms Cavallaro said.
Tributes from across the country continue to pour in for the Ms Maina, including condolences from New England MP Barnaby Joyce.
"She was an inspiration to many, and by all accounts, left a lasting impression on fellow students and staff here in the New England," Mr Joyce said.
Ms Maina was working as the head teacher at Nairobi's Hillcrest Preparatory School at the time of her death according to the school's website.
