Police are investigating whether speed and inexperience contributed to the incident at Hotham Heights.
Wangaratta highway patrol First Constable Shane Hughes said the man, 57, lost control of the bike about 4.15pm.
“He has come off the bike and hit another vehicle travelling the opposite way,” he said.
The man was flown to Essendon Airport where an ambulance then transported him to the Royal Melbourne Hospital.
Two highway patrol officers from Wangaratta and two Bright police officers attended the scene.
