The woman, 43, from Mulwala, was the sole occupant of her utility, which collided with a truck at the intersection of Spring Drive and Bull Plain Road about 3pm on Wednesday afternoon.
Emergency services worked for several hours to free the woman, who died while being flown to Melbourne for treatment.
The truck driver, a 35-year-old man, was uninjured but taken to hospital to undergo mandatory blood and urine testing.
Police are investigating the crash and will prepare a report for the coroner.
The woman is the second from Mulwala in the past month to die on the region’s roads.
On May 26, 66-year-old Mary Hidson died after the car she and her husband William, 69, were travelling in hit a tree stump along the Wangaratta-Yarrawonga Road, near Wangaratta.
The NSW toll is 154.
