Wodonga police said the b-double was travelling southbound on the Hume Highway when it veered onto the median strip about 2am.
Sergeant Amanda Meagher said the driver over-corrected, causing the truck to turn onto its side and spill its load.
The man, 49, suffered non-life threatening injuries and was airlifted to The Alfred hospital.
The southbound lanes of the highway have since been closed, with traffic diverted through Barnawartha, onto the Chiltern-Barnawartha Road, to Chiltern and back onto the freeway.
Emergency services remain on-scene, with delays expected.
Sergeant Meagher said police were investigating the incident but fatigue appeared to have been a factor.
Police are unable to say when the road might be reopened.
