A witness said there was a collision between a Ford Focus and Nissan 4WD at the intersection of the Hume Freeway and Old Barnawartha Road about 2.30pm.
The Patrol rolled five to six times and is now resting on its side near train tracks on the side of the freeway.
Two women have been pulled from the car but a man remains trapped at the scene.
The driver of the Ford is uninjured.
Northbound traffic has banked up and is being diverted onto Old Barnawartha Road.
It was initially suggested that train services may be delayed but The Border Mail has been unable to confirm this.
Police, paramedics, firefighters and rescue crews are on the scene.
