Transit police from NSW and Victoria checked trains and buses from 10.30am to 4.30pm with a labrador sniffer dog in operation.
The dog named Bear uncovered two men and a woman with cannabis.
An Adelaide XPT passenger, 36, was found to have a long-bladed knife after a search sparked by his anti-social behaviour.
NSW police transport regional commander Sergeant Michael Chubb said the man would be fined.
It was the third operation in 12 months and more are planned.
"The staff love it and the passengers love it, there's nothing worse than being stuck with somebody playing up," Sergeant Chubb said.
