Emergency services were called to the scene of the crash about 12.20am on Friday.
A southbound B-double left the road and travelled into the northbound lanes, hitting a car and tipping over.
The truck driver was airlifted to Melbourne with non-life threatening injuries and the car driver was taken to Wangaratta hospital.
Sergeant Colin Anderson said the crash, which occurred near Mansfield road, has sparked a large clean up operation.
“There are sweet potatoes everywhere,” he said.
“The freeway is still closed, it will stay closed until midday.
“It’s a mess.”
Police are looking at whether fatigue was a factor.
“The cause is still under investigation,” Sergeant Anderson said.
