St John, 22, will lead the club for the first time in Saturday's practice match against North Albury at Sandy Creek.
Taking over from Jydon Neagle, St John will play his 100th match for Wodonga Raiders in their round one match against Wangaratta.
Alex Daly has been promoted to deputy after enjoying a breakout 2018.
Raiders football manager Brad Smith said the club was thrilled with the appointments.
"Brad and Alex have come through the ranks and have a lot of respect at the club," Smith said.
"They were voted in by the players.
"They are only 22 and the future of the club."
The Raiders and North Albury will play a six-quarter match from 11am.
Meanwhile, Daniel Bond has signed with Ballarat league club Redan after moving to the region for university.
