Both clubs will wear specially designed jumpers to mark the occasion.
Rutherglen introduced the initiative in 2017 to honour the legacy of coach Karl Jacka who lost his battle with cancer the same year.
Co-coach Daniel Athanitis said it was a special day on the clubs calendar.
"It is a significant day and obviously close to the clubs heart," Athanitis said.
"Our president Greg Lumby works tirelessly to make the day a success and hopefully we can once again raise significant funds for the Albury-Wodonga cancer centre trust fund."
The inaugural year raised $93,000 with a further $13,000 last year.
The club will once again hold its high tea function in the club rooms.
Michelle Hensel is the guest speaker.
An auction will also be held along with a golf ball drop with $1000 up for grabs.
