Detective Inspector Winston Woodward said the armed robber walked into Lavington's Woolworths Caltex petrol station on Thursday at about 6.30am.
"The male there has demanded money from the female attendant, he's gone around behind the till, the female attendant has opened the till and the male has taken a sum of money," he said.
"The attendant did everything right. She opened the till as she was demanded to and hasn't tried to tackle this male in any way."
The man stole up to a few hundred dollars and ran away on foot along Griffith Road.
His face was covered by the ski mask, but he has been described as caucasian, 170-175 centimetres tall, and was wearing a dark blue hooded jumper with white stripes on the sleeve, grey tracksuit pants with white writing and a black backpack.
The weapon was a small silver knife.
Police will review CCTV footage and other forensic evidence from the scene.
"It's unfortunate it's happened, it's a terrifying crime, it's a serious crime and the detectives will put a serious amount of attention in relation to trying to identify the person," Inspector Woodward said.
"The attendant has been terrified in relation to this male producing a knife.
"It's been a while since we've had a robbery on a service station of this nature."
