There is a three-year board rotation process and current chairwoman Wendy Greiner is approaching the end of a six-year tenure in October.
MRT chief executive Mark Francis said Ms Greiner had helped guide the cross-border organisation.
"Wendy has seen a lot of growth and changes over the evolution," he said.
"We have a maximum of two terms by any board director.
"Our deputy chair will be running the selection process."
Deputy chair Greg Roberts said the board was looking for people with experience in corporate governance, strategy development, social media and stakeholder engagement.
"This is an exciting opportunity for experienced individuals to have a significant impact on an industry that is booming with award-winning tourism experiences," he said.

MRT works in partnership with Visit Victoria, Destination New South Wales, Destination Riverina Murray and providers in 13 local government areas.
"One of the key projects moving forward is the development of our Murray River Road project, getting people to experience the whole of the Murray," Mr Francis said.
The position of chair receives a small allowance to cover incidental costs and the director's positions are voluntary, however expenses are reimbursed for attendance at board meetings.
