NSW Chief Health Officer Dr Kerry Chant confirmed the new positive result during the Thursday morning press conference, but said the case has not yet been added to the state's tally.
Dr Chant said it is believed the family member had been isolating at home since the initial positive test last week.
The result comes as hundreds of Border residents lined up for more than an hour and a half to get their test in Lavington after border closures and public warnings encouraged those with symptoms including a sore throat, cough or fever to get tested.
A socially distanced line made its way from the Mirambeena Community Clinic mobile test site in Martha Mews, Lavington around the corner into Moore Street with Border residents heeding the advice of Dr Chant to get tested.
The increase of testing is hoped to identify any community transmission within the Albury-Wodonga area.
The Border Mail understands some of the people tested on Thursday had returned from Melbourne earlier in the week.

Murrumbidgee Local Health District, the service running the mobile COVID-19 clinic, didn't respond to specific questions put to them by The Border Mail on Thursday but said more than 500 people had been tested at the clinic since opening on Tuesday.
Dr Chant said the third family returned a positive preliminary result, with all three isolating under the care of a doctor.
"The index case did return from Melbourne and that is where we believe the virus was contracted," she said.
"I am very pleased with the turnout for testing in the Albury community and I would continue to ask all the border communities to be vigilant.
"Get tested even if you have the minimum of symptoms."
Glenroy man Tai Hunter was among the scores of people to attend for a test after having a cough.
He had to wait for an hour and a half from the time he joined the queue until he completed his test.
"MLHD continues to work with public transport providers to ensure passengers from Victoria travelling through and disembarking in our region are screened," a statement said.
More than 22,000 people in the local health district have been tested for the virus.
