Nicholson, who has played with the Patties since the under-12s, takes over at the helm from star opener Matt Crawshaw.
"It's a big honour and I'm thankful to be given the opportunity by the club," Nicholson, 28, said.
"I'm looking forward to getting into it.
"We've started training and I'm keen to build on last season's results."
Nicholson started playing first grade for St Patrick's in 2008-09 and made 198 runs in eight innings last season.
"Dean is focused on the challenge in front - among many things he will bring passion, pride, enthusiasm, enjoyment and that competitive edge we have spoken about as a group," club president Tony Maher said.
St Patrick's finished eighth last season with five victories and a tie with Neil Smith being the leading runscorer.
The provincial season is set to start on October 10 under coronavirus restrictions.
