He informed Tiger officials of his decision earlier this week.
Cook cited a promotion in his employment with Big River Roofing and the increasing demands of raising a young family as the major reason behind his decision.
Tom Anson has replaced Cook and will coach alongside Danny De Marte who was appointed co-coach in September last year.
In a major boost to the Tigers' premiership credentials, Cook has signalled his intentions to remain as a player.
Tigers president Chris Peirce praised Cook for making the tough decision and having the club's best interests at heart.
"Peter has got a promotion with his work which requires him to spend a fair amount of time away from home," Peirce said.
"To his credit, he was the first to admit that he couldn't allocate the time necessary to coach and approached the club about stepping down.
"Thankfully he is still keen to play and when fully fit, Cookie has proven himself to be one of the premier forwards in the competition.
"Cookie has done a tremendous job since he was appointed in 2018 and was instrumental in us making the grand final in 2019.
"He also did a power of work recruiting wise last year and steps down with a strong list that is capable of challenging for the flag again this year."

The Tigers finished third and runner-up under Cook before last season was ruined by COVID-19.
Anson was part of the Tigers most recent flag in 2013.
He also had a two year stint with Wodonga before returning to Barnawartha.
The Tigers have been able to keep their list intact since losing the grand final to Thurgoona by 71-points.
Defender Matt McLinden is the club's biggest loss.
They have added some class after signing Mitch Exton alongside Ben Kunkel and Jordan Hansted.
