It will be the first time a Tallangatta and District league match has been played on the Easter holiday since Thurgoona took on Dederang-Mt Beauty at Bunton Park in 2014.
"The clubs got together and we were able to come up with a date to suit everyone," Wahgunyah football operations manager Brett Skinner said.
"There was a cricket grand final clash involving players from both clubs and it went from there.
"It's a day where people relax and kick back and hopefully a good crowd gets along.
"Rutherglen and Wahgunyah have worked really well together in getting it off the ground."
Wahgunyah will host this year's match with Rutherglen to take charge in 2022 with donations to be made to the Good Friday appeal.

The clubs traditionally play each other in round one.
Rutherglen president Greg Lumby hopes the Good Friday match will develop into an annual fixture.
"We are looking forward to it," Lumby said.
"It's something we have spoken about as a club before and it's fantastic we have been able to get it off the ground this year with the support of the TDFL.
"Rutherglen and Wahgunyah always get a good crowd and I'm sure the support will be there again.
"With no football last year I'm sure people will want to come out for a look."
The Murray and Picola and District leagues have attracted big crowds to Good Friday matches over the past decade.
The crowd is presented capped at 1000 due to coronavirus restrictions.
Wodonga Raiders will take on Wangaratta at Birallee Park in a twilight match on Good Friday.
The TDFL season kicks off on March 27 with Beechworth playing Wodonga Saints, Chiltern meeting Barnawartha, Thurgoona taking on KSC, Tallangatta doing battle with Mitta United and Yackandandah hosting Dederang-Mt Beauty.
The Hume league gets underway on April 10.
